Belknap was born in Damariscotta. Parents include David Belknap and Louise Belknap. She studied at Lincoln Academy, Newcastle Maine. While attending Middlebury College in Vermont she began to develop an interest in performing. The next place she visited was the American Conservatory Theater where she earned a Masters of Fine Arts. The credits on her TV show include regular series roles as a part of Medical Investigation and The Handler (related to ER and Third Watch), a frequent appearance in Deadline and guest-starring parts on Law & Order Law & Order Special Victims' Unit as well as Without a Trace. Belknap played in Alchemy. Her feature film debut was in The Reality Trap. Belknap joined the cast of the crime drama that has been running for decades CSI: NY as Detective Lindsay Monroe in 2005 during the second season's Episode Zoo York.
